Biochemistry

Biochemical testing needs be done under standard conditions – in the morning on an empty stomach without having taken any liquids for up to 10 hours (after 12 hours of starvation during the night) or at least 2 hours after the last meal and 24 hours exclusion of alcohol intake. It is also needed to avoid the use of coffee and cigarettes before sampling.

Urine for urine culture

  • Appropriate for testing is the intermediate portion of the first morning urine or, exceptionally, urine released at least three hours after the last urination.
  • Thoroughly wash your external genitalia with water and soap.
  • The first 20-25 ml urine is allowed to leak into the toilet bowl.
  • The intermediate urine portion is collected in a sterile container. Required quantity – 5 ml.
  • Do not touch the container or its lid on the inside with hands/clothes/towel.
  • Close the container gently to prevent material from leaking. Leaking containers are not accepted for testing!
  • The urine is taken to the laboratory for examination within two hours of its excretion.

Sputum of microbiological examination

  • It is advisable to test sputum, which is released in the morning upon deep expectoration.
  • Do not use solvents or toothpaste as they contain a disinfectant.
  • Deep breathing and coughing is required several times before obtaining sputum suitable for testing.
  • The sputum is released directly into a sterile container with a wide opening, with its edge pressed against the bottom of the lower lip so that to collect the entire amount.
  • Close the container gently to prevent material from leaking. Leaking containers are not accepted for testing!
  • The sample is taken to the lab within two hours of expectoration!

Feces for microbiological examination

  • Feces for microbiological examination is placed in a sterile container with a wide opening.
  • Avoid contacting the sample with urine.
  • Required quantity: about 1g. (pea grain size) or > 2 ml. (in case of liquid stools)
  • Close the container gently to prevent material from leaking. Leaking containers are not accepted!
  • The sample is delivered to the laboratory within two hours of its excretion.
  • Samples can be stored for 24 hours in a refrigerator at 4-6 degrees Celsius before being taken for testing.

Throat secretion

  • The secretion is taken with a sterile swab in the morning on an empty stomach before teeth washing. Do not take liquids.
  • The tongue is pressed down with a spatula. Sweep with the swab the two tonsils, the back pharyngeal wall, each polyp, redness, swelling or ulceration.
  • The swab is placed in a transport environment in order to preserve the viability of the bacteria.
  • It is to be taken to the laboratory for testing.
  • The secretion may be taken on-site at the lab up to 10 a.m.

Wound secretion

  • In case of surface secreting wounds, material is taken by aspiration with a sterile syringe.
  • Exceptionally, material is taken with a sterile swab.
  • In case of an abscess/phlegmone incision or puncture is made and the material is aspirated with a sterile syringe.
  • It is placed in a transport environment.
  • The material is taken to the laboratory for testing.

Nasal secretion

  • The secretion is taken with a sterile swab
  • Do not use medications for local use at least 24 hours
  • The swab is placed in a transport environment in order to preserve the viability of the bacteria
  • Secretion can be taken on-site in the laboratory

Cervical secretion

  • Secretion is taken by a gynecologist and is taken for testing at the lab.

Vaginal secretion

  • Secretion is taken by a gynecologist and is taken for testing at the lab.

Prostate secretion

  • Secretion is taken by a urologist.
  • Collect in a sterile container.
  • The secretion is taken for testing at the lab.

Urethral secretion

  • Secretion is taken to the laboratory between 9 a. m. and 4 p.m.

Ejaculate for microbiological examination

  • Thoroughly wash your genitalia with water and soap.
  • The ejaculate is separated directly into a sterile container.
  • Close the container gently to prevent material from leaking. Leaking containers are not accepted for testing!
  • The sample is taken to the lab within two hours of separation.

Taking a perianal sample with special Scotch tape

  • Sample – Scotch tape sample is taken in the morning, without washing with water, before defecation, so that there are no stools on the tape.
  • The Scotch tape is detached from the slide and sticks to the anus.
  • Carefully grasp the tape at its end and stick back onto the slide.
  • The material is taken to the laboratory for testing.

Drainage secretion

  • The end of the drain tube is cleaned with an antiseptic solution.
  • Allow some exudate to drain off, then collect 2-3 ml. from the secretion into a sterile container.
  • Close the container gently to prevent material from leaking. Leaking containers are not accepted for testing!
  • Not suitable for anaerobic culture!
  • The secretion is taken to the laboratory for testing.

PCR (DNA analysis)

  • For men, the material is taken on-site in the laboratory between 9 a. m. and 4 p.m.
  • For women, the material is taken by a gynecologist and is taken to the laboratory for testing within 1 hour after being sampled.

Breast milk for microbiological examination

  • Thoroughly wash your breast with water and soap.
  • Squeeze 1-2 ml. for cleansing of the canals from the cutaneous microflora.
  • Required quantity for testing: > 2 ml. directly segregated into a sterile container.
  • Close the container gently to prevent material from leaking. Leaking containers are not accepted for testing!
  • The sample is taken to the lab within two hours of separation.

Urethral secretion

  • Secretion is taken by a urologist.
